低劑量照射促進干細胞內(nèi)吞熒光納米藥物載體的定量分析研究

摘要:低劑量電離輻射(LDR)能夠促進干細胞的增殖和細胞因子表達等生物學(xué)行為,然而LDR對調(diào)控干細胞攝取納米顆粒的行為仍有待闡明。本文通過復(fù)乳法制備的擔(dān)載近紅外熒光素吲哚菁綠(ICG)的熒光納米藥物載體(ICG- NPs),具有優(yōu)異的分散穩(wěn)定性和生物相容性。將人臍帶間充質(zhì)干細胞(MSCs)進行LDR照射后,與ICG-NPs共孵育,制備ICG-NPs標記的干細胞。研究發(fā)現(xiàn),不同吸收劑量的LDR暴露可以促進間充質(zhì)干細胞對納米載體的內(nèi)吞作用,在同等條件下具有更高的干細胞標記率,證明LDR可以提高干細胞對納米載體的內(nèi)吞效率和標記的時效性。此外,低劑量照射還促進了干細胞的增殖和細胞干性特征的維持,因此LDR協(xié)同納米載體處理干細胞,為干細胞疾病治療過程中長效示蹤與行為調(diào)控提供了新的技術(shù)方法。

Low dose Radiation Enhanced Uptake of Drug-loaded Fluorescent Nanocarriers in Mesenchymal Stem Cells

Abstract:Low-dose ionizing radiation (LDR) can promote biological behaviors of stem cells, such as proliferation and cytokine expression; however, its effect on nanoparticle uptake by stem cells is still a lack of report. Herein, the near-infrared fluorescent nanocarriers loaded with indocyanine green (ICG-NPs), fabricated via the double emulsion method, exhibited excellent suspension stability and biocompatibility. Human umbilical cord mesenchymal stem cells (MSCs) were irradiated with LDR and subsequently co-incubated with ICG-NPs to prepare ICG-NP-labeled stem cells. The results revealed that LDR exposure at varying absorbed doses enhanced the MSCs endocytosis of nanocarriers, resulting in higher cellular labeling efficiency under identical conditions. These findings demonstrated that LDR significantly enhanced the endocytic efficiency of stem cells toward nanocarriers while reducing the time required for effective labeling. Furthermore, low-dose irradiation promoted MSC proliferation and maintained the stemness. Thus, the combined strategy of LDR and nanocarriers offers a novel technical platform for long-term tracking and modulating of stem cell behaviors in therapeutic applications.
