摘要:目的:探究穩定無poly(A)尾RNA的方法。方法:構建有潛在穩定作用的RNA結合蛋白體系,如LicT-RAT、MCP-MS2體系,通過切除靶基因的poly(A)模擬天然無poly(A)的RNA,研究這兩種體系對RNA的穩定和激活作用。結果:所構建的兩種蛋白結合適配體體系均能很好地穩定無poly(A)尾mRNA并促進靶基因的翻譯表達。結論:本文成功構建LicT-RAT和MCP-MS2兩種穩定RNA的體系,有較好的應用潛能。

Abstract:Objective: To explore the method of stable poly (A) -tailed RNA. Methods: Construct RNA-binding protein systems with potential stabilizing effects, such as LicT-RAT and MCP-MS2 systems. Simulate natural poly (A) -free RNA by resecting the poly (A) of the target gene, and study the stabilizing and activating effects of these two systems on RNA. Result: Both of the two constructed protein-binding aptamer systems can well stabilize poly (A) tail-free mRNA and promote the translation and expression of target genes. Conclusion: In this paper, two stable RNA systems, LicT-RAT and MCP-MS2, were successfully constructed, which have good application potential.
